Alright everyone, welcome to another episode of Sunday Science, the Go on Pro podcast. Our Sunday Science episodes hope to bring you some information from a few different studies, very recent studies. And today we're going to be talking about microplastics and forever chemicals and inflammation going on in the body either, um, from aging perspective or from the in utero perspective and later on childhood inflammation of the colon.

Okay, so there's mounting evidence that there are nanoplastics, microplastics, and forever chemicals, the perfluoroalkyl compounds, okay? These compounds out there affecting a multitude of organ systems when you ingest them. Um, and they could get exposure to your mucous membranes, to your skin, and potentially enter there, and could be aerosolized from your clothes into the air and then breathed into the lungs, and um the use of plastics has exploded in the last 20 years.

uh we're only beginning to root out the causative plastic compounds and some of these plastics are termed food safe. It's okay anywhere to put any food compound into these plastic compound, let's say containers, et cetera. Some of them are heated at least to a certain degree and those plastics are termed food safe. That could be a misnomer. We don't know yet at this point because we don't know exactly where and which ones of these microplastics and nanoplastics come from which plastics out there that we have.

Okay, so today's episode, we're going to explore three different articles that have come out in the last few months showing how forever chemicals and microplastics affect inflammation in the intestines and the liver and the lung and increase aging and senescence of cells. And one part of one of the studies actually showed increased tumorigenesis in the colon after high normal exposure. When I say high normal exposure, we're talking the doses that they gave to these individual mice was the same exposure that can be found in human feces. Okay?

so human feces normal and this is a high normal but still showed tumorigenesis, okay? So then there was uh this other study, the first study we're going to talk about is talking about forever chemicals found at birth in either cord blood or in the dried blood spots uh from the neonate at birth or from maternal serum um in these patients. And then later developed inflammation in the child.

This is the child between one and eleven years after childbirth showing that that exposure is associated with inflammatory conditions, okay? Certainly, there's an overabundance right now already of inflammatory conditions in the human spirit right now, right? An increase in GI disturbances, leaky gut. We know that inflammation can lead to the aging phenotype in a multitude of cells, multitude of organs, from dementia to cardiovascular disease to diabetes, uh NAFLD, the fatty liver disease.

to inflammatory bowel disease, to irritable bowel syndrome, and your endocrine system, your hormone system as well, okay? So, uh, let's start with the study, uh, on forever chemicals, okay? So, this was a study, um, by Majid Jouina et al. It was in the Environmental Pollution

Journal, ingestion of human-relevant mixture of environmentally sourced microplastics promotes inflammation and tumorigenesis in the mouse colon. So, here's what they did. They conducted an untargeted perfluoroalkyl compound analysis in early life samples from three cohorts. So the first cohort they had dried blood spots from offspring at or near childbirth, and then there was another cohort that had cord blood from two prospective birth cohorts with and without inflammatory bowel disease in New York.

And then there was another fourteen samples of maternal serum during pregnancy from a birth cohort in Mexico City. Then, sometime between age one and eleven, one cohort had between age one and six, the other one between six and eleven.

and they took fecal calprotectin levels of these kids, okay? And correlated the levels of the perfluoral alkyl compounds in these mothers and at the children at birth and was associated with calprotectin elevation later. Now, is this causative? It is, there was some evidence there that it might be causative, but that's not a hundred percent, right?

So just because you are ingesting a lot of polyfluoroalkyl compounds and getting into the bloodstream of your child or into your bloodstream when you're pregnant does not necessarily mean that that's the cause. What if the cause is actually the fact that you're eating a lot of fast food and that's why you had that number of polyfluoroalkyl compounds or you were living in a particular location that was unhealthy, you were being unhealthy at the same time and then that could um subsequently lead to an issue. But this is further evidence.

We already have some evidence in humans that it is causative for, um, calprotectin and and and, um, and polyfluoral compounds, showing that there is inflammation in adults associated with calprotectin associated with inflammatory bowel disease or irritable bowel syndrome. And now we've shown potentially that same causality or association between polyfluoral alkyl compounds. Okay.

So, the and that beta um covariate adjusted beta estimate was .44. That means for every decile increase in

polyfluoral exposure, there was an equal increase in exposure approximately a point four four exposure to calprotectin in the child, okay? So, um it's not only it's somewhat dose related and it's also dose related in this study, they actually um took a pooled sample of all the perfluoral alkyl compounds.

and found that they were the sum was associated with the increased calprotectin, but also if they teased that out, the more um what they call legacy or uh the previously banned perfluoroalkyl compounds PFAS, um and um PFOS, those levels were even more correlated with the levels. So, uh here we have calprotectin levels increased. Okay, so what is calprotectin?

Calprotectin is a protein released by neutrophils into the lumen of your intestines if you have certain inflammatory conditions. So upon recruitment of neutrophils, neutrophils are inflammatory infection-fighting or inflammatory cells in the body that are a part of your immune system. Neutrophils are recruited to go to spaces where they are concerned, the body is concerned about invasion of either a foreign substance or a foreign organism. Okay.

so, those neutrophils, when they get to the mucosa, to where they were recruited to go, some signal from the body from that area said, "Hey, we need your help." There is leaky gut. There is uh the...

Um in leaky gut you have translocation across the membrane of bacterial cells and also of bacterial compounds called lipopolysaccharide.

lipopolysaccharide causes an increase in inflammation. So upon getting to that area, those cells secrete this compound called calprotectin. So it's released into the lumen of the intestines upon recruitment to that mucosa. It's been associated with inflammatory bowel disease in adults. Mothers with inflammatory bowel disease have kids with elevated calprotectin until the kids are about three years old, but this is the first time this has been really associated.

there's been a few studies with calprotectin associated with polyfluoroalkyl compounds in the perinatal period, okay? So mothers, those offspring also that have that calprotectin and are born to a mother with inflammatory bowel disease, they have loss of intestinal microbial diversity and immune dysregulation at least for the first part of their life.

And this is the first time this has shown calprotectin levels increased later in patients with perfluoroalkyl compound

secretion and and absorption, okay. So, um they also as part of this they teased out the patients with with and without maternal inflammatory bowel disease. So inflammatory bowel disease is either Crohn's disease or ulcerative colitis. This is more serious than your just usual inflammatory bowel syndrome, okay. Or sorry, irritable bowel syndrome. This is inflammatory bowel disease. Okay, so, um with those with maternal inflammatory bowel disease,

were more likely to have higher perfluoro-1-octane sulfonamide, so that's the PFOA, and also PFFP, um alkyl alkylate levels in dried spots, and respectively those were among the top contributors to offspring calprotectin, okay? So, um, so here we are. So, what does that mean? Uh, could it be that that this is part of the cause of inflammatory bowel disease and or irritable bowel syndrome? Some of that inflammation.

we do know that there is a difference in the microbiota of the intestines, and we're going to show a study here in a little bit, not necessarily perfluorooctane compounds, but also microplastics and nanoplastics. So these are, this is kind of the same pathway we'll show that there is senescence, but also a difference in the microbiota.

so microbiota of the mother and potential inflammation that's being exchanged and transmitted to the child, inflammation and that those compounds getting there to cause fecal calprotectin up to 11 years after birth. Okay, so again, not necessarily causative, but we're getting there.

I think that it could be at least part of the story. And certainly the abundance of associated with the perfluoroalkyl compounds, a highly processed diet, boom, you have either one of those or both causing this inflammation in the child. Okay, so, all right, so let's go to our second article here.

So, our second article, um, suggests that, um, all right, so we're looking at this article that I was just telling you about a little bit ago, um, and microplastics. Okay, so, all right, so the in this study they took mice and they gave them a human-relevant mixture of environmentally sourced microplastics and it it showed that it promoted inflam-

This is in the journal um Environmental Pollution.

Right. And the last article by the way was Vishal Midya, um, and that was in Clinical Gastroenterology and Hepatology in 2026. All right, so this article shows that there is an increasing, um, consumption of plastic and those microplastics at doses, same doses that could be a human could be exposed to, 50 to 500 or sorry five to five hundred, um, micrograms per gram into the intestines of these mice, okay?

so, um, and that reproduces the portion proportion that we find in human stool. And that 500 is a pretty high level, but not unheard of, okay? So, at that high level, they gave a 30-day exposure to that microplastic exposure with dose-dependent, and that showed, um, a dose-dependent alteration in colonic transcriptome, down regulation of heat shock proteins, T-cell activation, cytokine receptor interaction signaling pathways, and those, uh, signaling pathways

um were showed lymph node dysfunction as well. And also in a colonic inflammation that was artificially introduced. So they gave DSS also to the mouse at the end of the study after this, and those mice develop a colitis, an inflammation of the colon. After this microplastic dosing that

colitis or inflammation of the colon was increased in those that received the microplastics compared to those that didn't. And, um, a 75-day exposure to the high level at that 500 micrograms per gram showed an induction of tumorigenesis. Okay

So same, uh, patients are are given the colitis by DSS and colitis is somewhat tumorigenic in and of itself, but that tumorigenesis is increased in those patients that had the higher dose of the microplastic dosing. Okay, so there was endoscopic, macroscopic, histologic evidence of tumorigenesis, increased something called cyclin D1, MYC protein. These are tumorigenic compounds that are found in the mice that were exposed to that.

but not as much in the mice that had the colitis induced without the microplastics, okay? So potentially here, you have already um a patient with, this is a mouse study obviously, but let's say theoretically you have a patient with uh developing cancer or they have a developing inflammation of the colon such as ulcerative colitis. If they're exposed to more microplastics, they might be more likely to develop tumors.

We do know that patients with ulcerative colitis are prone to developing colon cancer, um, at a little bit higher rate than persons without colitis or ulcerative colitis.

So, um, the... um...
Dr. Brent Laartz
That environmentally sourced microplastic mixture is the same kinds of microplastics that you and I may be ingesting every day, okay? So, this is something that is very disturbing, okay? So, the consumption of these mice and a lot of these studies, you think, oh, it's just mice and they're in they're giving them super physiologic doses of plastics, right? Or something like that. This is the compounds at the same concentration that they can find in human stool, okay?

So, again, this is an interesting study. All right, so the third study we're going to talk about is polystyrene, okay? So, um they can make these uh things called polystyrene nanoparticles. And polystyrene, as you know, is very abundant in our food containers.

that we get for coffee, for a lot of different things, all right? So, um, and those plastic particles size less than a thousand nanometer represent the final products of plastic degradation that are in potentially the water after polystyrene is broken down. Okay. Then they're now widespread in water, air, soil, and therefore if they're on soil, they're being absorbed potentially onto and into plastics, uh, of inside microplastics and nanoplastics, into your plants that you might ingest. Okay.

so, um then they can get into from there into animals that you might ingest, such as cows, pigs, chickens. Okay, so if they're in your corn, if they're in your soybeans, if they're in the grass, then they can be in the animals and the food that you consume. So, and these potentially can accumulate in the intestines, liver, lungs, and can potentially threaten human health, okay? So the physical presence may not be the only problem, okay?

So these nanoplastics, when they get into your bloodstream or any other tissue, to mucosa, to into others of your body, they may actually be a harbor or a vector for other environmental contaminants. Heavy metals have been associated with these and persistent organic pollutants such as, um, the forever chemicals and or the, uh, pesticides and or, uh, pathogenic microorganisms could gain their way into the body by latching themselves onto these microplastics and nano- nanoplastics, okay? So, and that leads to combined pollution effects.

Um, alright. So, here we have now this study, they took these polystyrene nanoparticles and they and so let's let's talk about the article, let's give you the article.

polystyrene nanoplastics promote inflammation and aging in young mice through the oral gut microbiome axis. This is in Frontiers of Immunology. Again, I'm not going to be showing any of the um of the article here and I'm not going to be showing any of the the graphs and the and the graphics. Um, I I the plas the graphics in these articles were pretty busy and pretty small and would be very difficult for people to uh find uh them and be able to read them in on this uh um small format.

Bzzzz So, all right, so

this article wants to suggest that we can find and monitor the aging process and see if it's affected by these polystyrene nanoparticles, okay? So, um these particles uh average particle size around 91 nanometers, um associated for the largest proportion and so what they did in these

mice was a expose the mice to the nanoparticles and they were able to show um that there was a multi-tissue inflammatory response in these young mice. So, they took young mice and they're not supposed to have any aging compounds or any inflammatory signals in the body because they're normal healthy mice, right?

so, but they found um that there was an induction of something called the p38 MAPK MAPK pathway. And that's one of the key pathways in regulating cellular senescence. When you phosphorylate uh these um this p38, it leads to pro-inflammatory inflammatory markers such as IL-6, TNF alpha, there's also IL-1 beta, um and p16 and p21. These are the pathway for senescence and aging.

Okay, so the so if you can show that there is phosphorylation of p38, you can show that there is this pathway being activated, okay? So they demonstrated a significant elevation in that phosphorylation level in the lung and the liver. So now you've got now showing that you've exposed these mice to the polystyrene nanoparticles and and basically fed for six months, so they were given ad libitum diet before the subsequent experiments.

Okay, so and then they gave them the uh diet that was rich in the uh polystyrenes for about uh six months. So two months of normal diet, two months to eight months. So these are very young mice and you can see uh that there was an increase in phosphorylation in the MAPK p38 pathway, okay? So that was already the start of it, okay? So that's the start of the inflammatory promotion heading off into senescence, okay? So now they then also...

found these two proteins called p21 and p16. These are cell cycle arrest core proteins. Um, and they showed that, um, a significant upregulation of those proteins in the lung and liver also. So here we've got the p38 phosphorylated, and then you've got cell cycle arrest, and then you've got senescence because of the p21 and p16. And so those quantitative PCR results show that those genes were upregulated as well.

another part of the study, the next part part of the study is that then what you would want in a person or a mouse who is developing this senescent cells and developing some some pathways where these proteins within the cell are causing the cell to age faster, you would want to try to arrest that, right? You'd want to try to make

that better

and how do you do that with antioxidants, right? Your body has an antioxidant and a detoxification pathway that could help to mitigate that effect. But they also showed in the same study that the same mice also had an impact on mRNA expression levels of antioxidant genes and detoxification genes. These are um some enzymes called POR and CYP8B1 and GSTA1, GSTA2. These and then there's also a transporter MDR3.

these compounds would normally transport some of these toxins, some of these um senescence active pathway proteins out of the cells and and or into some degradation pathways so that they could get rid of it out of the cell, but that was down regulated and the detoxification and antioxidant enzymes were de-de-regulated or down regulated in the cell, suggesting now you have some multiple genes involved here from the pathways of senescence. Again.

the next part of this that they also did because they could is they could also examine the microbiota also, which could be part of this senescence pathway and signaling, okay? So they then took these mice that they gave those same polystyrene nanoparticles to and they examined their microbiota in both their mouth, in the oral cavity, and their intestines. And the fecal samples, um, and the oral samples showed that there was a basically what's called a dysbiosis.

It showed a significant reshaping of those from the abundance of proteobacteria, um from and the were increased in firmicutes, which are good bacteria, were inhibited. Um and there was a decrease in lactobacillus, decrease in Akkermansia, um and there was an increase in coronabacterium and streptococcus, which potentially could be non-beneficial bacteria, okay.

so beneficial bacteria are decreased and some of the non-beneficial bacteria are increased in the patient, not patient, in the mice. Um, I'm a I'm not a researcher in the lab, so I'm not used to saying mice, but the patients that were the mice in this study, um, had this exhibited, okay? Same thing in the intestinal microbiota, you had the similar process, and um, there was a significant difference in the bacteria, okay?

So, so now you've got uh from the intestines and the issues with the microbiota being adjusted and altered by this polystyrene, which is abundant in the soil, air, and everywhere.

and we have then senescence increasing, phosphorylation increasing of the senescent cells and then the increase of senescence markers and then also then a decrease in the amount of antioxidant and detoxification genes that would normally be upregulated in this horrible situation of being a uh being

invaded by these polystyrene nanoparticles. But those are downregulated. So normally you would want those there, right? So now you've got those decreased, okay? So a few different studies here, not necessarily all interrelated exactly, but three small studies just in the past two months showing forever chemicals in our kids after being

exposed in utero and microplastics ingested um by the mouse increasing inflammation in the intestines and then tumorigenesis. Okay.

microplastics being found, uh being given to mice and this polystyrene promoting um the senescence of cells and aging and markers of aging of cells and then in in inducing detox and antioxidant dysfunction and downregulation of those genes that would normally try to prevent what is happening in your body. Okay, so three three great articles, three interesting articles.

And I'll tell you, um if you if these kind of studies don't make you want to get rid of completely your plastics in your household, in your clothes, because, you know, even um I'm wearing a probably a synthetic compounds shirt, but I am wearing cotton t-shirt, uh I am wearing cotton underwear, I'm wearing cotton socks.

I'm wearing as much as I can. I'm trying to decrease my plastic usage. I have done 90% of my life deplasticized, okay? So that is being done. If that if this

kind of study showing inflammation in your intestines doesn't make you want to change and get rid of plastics in your life? I don't know what would. And get rid of also ultra-processed foods because the vehicle for the plastics that are in your life are not only just these other compounds that you're wearing, you're using, and all of that kind of thing, it's in the food that you're eating. Okay, so forever chemicals, nanoplastics in our food. Unless you can somehow get rid of them from now the food chain, right?

So, um, all of this inflammation is highly related to disease processes, disease processes from inflammation in your intestines, which can cause mental ill health issues, can cause cardiovascular issues, and can cause endocrine issues and stress hormones increase. There's such a big increase in the thought process of trying to decrease cortisol by doing fad things, right? But the fad thing should be getting rid of microplastics and getting and taking care of the microbiome by getting rid of microplastics and ultra-processed foods. Going to whole foods, okay?

so that kind of thing is where we need to be. Where we need to be for the future. utensils that you use in the kitchen, anything that you use to store your food, any restaurant that you go to that's going to produce your food for you if you go out to eat a lot, those kind of things are the are the potential areas where you can really make a difference. But then the next 10%, 20%, we don't even know exactly what percentage that is, is everything else you can do. Okay, so every time there are studies where they've shown

that certain clothes that have certain microplastics that are made of certain plastic compounds actually for a certain time period after you buy them new and it's not just the first wash, it is for quite some time that each time you wear them and and maybe even sweat in them that they give off microplastics, nanoplastics into the air, okay? So, all right, so let's um go a couple of steps further here and um let's talk about what we're doing in the future.

Also, um, we're going to show some different studies uh with intrauterine as well. We'll try to incorporate that because I think that's an important place to think about this. And uh there was a study that showed that fertility was increased by decreasing microplastic and ultra-processed foods out of people's lives. There's a multitude of studies showing that your microbiome is important.

Obviously that's important to me, uh, is the microbiome because that is where, um, we have the most evidence that there is a direct connection to your health, okay? So microbiome affects everything.
